I use Simplify3D, on the G-code tab it allows me to enter extruder offsets. Do I still need to enter G10 in the config too? How can I verify that the offsets are accurate in either situation? 3 extruders I use Ex2 as 0

I suggest you enter the offsets in the G10 command and do not enter them in S3D. But you can do it the other way round if you prefer.
