Can you power the Prusamendel 2 only through an USB cable hooked up to the computer? Or do you need a power supply?

You need a power supply. Something to provide the 12v

The 5v from usb only powers the logic level chips.

Steppers, hotends and heated beds all require external power, normally 12v.
