Pronterface - How to start Printing?

I have problem with start to printing sad smiley My config: Sanguinololu 1.3a, Pronterface, Slic3r, Prusa Mendel, Sprinter Firmware
Manual mooving motors work well, positioning too
After loading STL everything works good. Gcode is generated. When I load gcode and hit "Print" nothing happened - where is the problem? Please help

How long do you wait?
Do you preheat the extruder/bed to temperature before printing?
